As a precursor of NAD+, NADH, NADP+, and NADPH, niacinamide plays an integral role in cellular metabolism, cellular proliferation, and the repair of damaged cells. In addition to supporting a healthy metabolism, niacinamide serves the vital function of regenerating the body’s master antioxidant and detoxifying molecule, glutathione.
So, what makes this particular form of vitamin B3 so unique? To answer that question, we need to look more closely at the other forms currently available.
There are four forms of vitamin B3 currently offered in nutritional supplements:
- nicotinic acid – a natural, “acid” form of vitamin B3. Possesses mild antioxidant, anti-inflammatory properties (i.e. weaker than niacinamide); in doses greater than 20 mg can produce an uncomfortable red, flush covering the entire body, which lasts from 20-60 minutes. For this reason, it’s usually not used in multivitamins as a source of vitamin B3.
- niacinamide (nicotinamide) – a natural, “basic” form of vitamin B3. Possesses powerful antioxidant, anti-inflammatory properties. The most potent antioxidant of the four forms of vitamin B3; more powerful than vitamins C and E. The form most commonly used in multivitamin supplements.
- nicotinamide riboside – a natural, non-flushing form of vitamin B3. A newer, more expensive form of vitamin B3 touted as a superior precursor to NAD with questionable advantages.
- inositol hexanicotinate – a natural, non-flushing form of vitamin B3. Possesses 30% lower absorption than other forms of niacin.

NIACINAMIDE – A POWERFUL PROTECTIVE SHIELD
As a powerful antioxidant, niacinamide protects your cells against harmful reactive oxygen species (ROS) – also known as “free radicals”. These highly reactive molecules can damage delicate cellular structures and are primary drivers in the body’s aging process. In excess, free radicals accelerate the natural aging process.
A perfect example of free-radical damage is a sunburn, where the UV rays of the sun generate more free-radicals on the skin than the body can effectively neutralize – with antioxidants – resulting in a transient burn. Wrinkling of the skin is another effect of cumulative free-radical damage, which can be increased by free-radical generators such as excessive sun exposure, air pollution, and heavy smoking and alcohol consumption. Put them altogether without adequate antioxidant protection (e.g. plant-based diet, organic juices, nutritional antioxidants such as vitamin A, C, E, carotenes, copper, selenium, zinc) and you have a recipe for looking older than your years.
In addition to accelerating the aging process, unopposed free-radical generation can lead to cellular dysfunction and increases the risk of a number of diseases including depression, chronic fatigue, cognitive impairment, mitochondrial damage and dysfunction, metabolic syndrome, neurodegenerative diseases such as Alzheimer’s and Parkinson’s disease, atherosclerosis, and diabetes.
NEUROTROPHIC AND NEUROPROTECIVE EFFECTS
Niacinamide is neurotrophic, which is to say, it supports the health, growth, and repair of all cells in the brain and nervous system. Through its innate antioxidant properties, and ability to regenerate the body’s master antioxidant, glutathione, niacinamide functions as a powerful antioxidant. Coupled with its role in mitochondrial energy production, and a wide variety of cell nourishing and sustaining activities, niacinamide also possesses powerful neuroprotective activity.
Like other neurotrophic vitamins and minerals (e.g. vitamin C, zinc) whose intake is often suboptimal or deficient due to a variety of factors including nutrient poor diets, excess refined sugar, increased requirements due to stress, and medication induced deficiencies – niacinamide in optimal amounts promotes the formation of new neurons (i.e. neurogenesis).
